Purification And Partial Biological Functions Of Body Wall Collagen From Asterias Rollestoni

The Asterias Rollestoni, a kind of widely distributed neritic Asteroidea, belongs to Echinodermata phylum, whose body wall is composed of mainly collagen, the most common protein in many animal bodies. Collagen is a major component of extracellular matrix (ECM), and has a unique three-dimensional structure with a unique amino acid composition. Owing to wonderful properties of its low-antigenicity, easy-biodegradability and excellent biocompatibility, collagen is used more and more widely in areas of biological material development, food industry, cosmetic manufacture, and medical production as well.Recently, there have been many interests in establishing efficient techniques to isolate collagen from skin of fish, and skin, cartilage and so on from mammals, and to apply them in development of various kinds of biological materials. However, there has been no report on collagen purification from the body walls of starfish Asterias Rollestoni and its biological functions on cells in vitro.To estabilish the methods to isolate and purify collagen from starfish body wall for utilizing the abundant starfish resources, reveal its biological functions on corneal cells, skin fibroblasts and tumour cells in vitro, investigate the feasibility of its practical application in clinic ophthalmology, cosmetic manufacture and tumor research, the collagen from starfish's body wall was extracted by reformative enzymatic extraction, and its biological functions on cultured corneal cells, skin fibroblasts and tumour cells was studied. The findings of this study not only lay important theoretical and technical foundations for fabrication of corneal tissue engineering scaffolds, manu- facture of cosmetic and research of anti-tumor, but also have significant scientific value and economical meanings.Acidic extraction, neutral salt extraction and enzymatic extraction was used to extract collagen from the body walls of starfish .The result showed that the yield of collagen by enzymatic method was highest, so this method was chose to prepare collagen in this article.
