The tree branch of Lauraceae which holds the balance in angiosperm class system is a big wood class group.However at present the evolution relationship about this group is extremely deficient, this made the resources cut down sharply because of excessive utilization on it for a long time,even some of them are in severe danger.And now the limited understanding about the endangered mechanism and genetic diversity has extremely affected the protect to these endangered plants.Thereby carry on systemic classify and genetic diversity analysis on Lauraceae plants has important academic and applied value.Cinnamomun micranthum Hayata is an important cash plant,which oil is the essential refined material,which wood is paper making material and with beautiful shape is gardens and road plant,its distribution is disconnected between Chinese mainland and Taiwan.Excessive utilization on Cinnamomun micranthum Hayata resources for a long time, this made the resource cut down sharply.Now Cinnamomun micranthum Hayata is endangered, there is only one bigger distribution of Cinnamomun micranthum Hayata forest in the natural protect reserve of Wanmulin,Jianou,Fujian,it's one of the state-protected three-grade and endangered plants.Aimed at the study of Lauraceae system classification in domestic and overseas,genetic diversity and genetic variation between Cinnamomun micranthum Hayata natural populations and Ex-situ conservation poputations and relationship among Its Close Relative Species of Cinnamomum camphora (L) Presl,Cinnamomum burmannii(C.G.etTh.Nee) Bl,Phoebe bournei(Hemsl.) Yang. Litsea subcoriacea Yang et P.H.Huan. Neolitsea aurata(Hay.) Koidz,Lindera communis Hemsl and Persea Americana Mill.were studied by inter-simple sequence repeats(ISSR) molecular marker.The research aimed at genetic variation between Cinnamomun micranthum Hayata populations,so that can offer the molecular theoretic and practical method for conservation and utilization of Cinnamomun micranthum Hayata populations.The results are as follows:(1)The DNA extraction of Lauraceae plants has stabled.This is very difficulty because of the high level of secondary compounds and their diffent distributing,or example,pigment,amylase and hydroxybenzene.The improved CTAB method which is suited for Cinnamomun micranthum Hayata and Its Close Relative Species of Cinnamomum camphora(L) Pres.,Cinnamomum burmannii(C.G.etTh.Nee) Bl,Phoebe bournei(Hemsl.) Yang.Litsea subcoriacea Yang et P.H.Huang.Neolitsea aurata(Hay.)Koidz,Lindera communis Hemsl and Persea Americana Mill is put forward in this study,but the SDS method of the DNA extraction for Cinnamomum burmannii is used before pretreatment.The extracted DNA is suitable for ISSR-PCR.(2)A optimal reaction system suitable for the ISSR analysis in Cinnamomun micranthum...
