With the entrance of X-nomics era, new experimental technologies and scientific ideas provide opportunities to the progress of classic subject. The incorporation of the bioinformatics tools and sophisticated experiment technologies enables researchers to establish their hypothesis more globally and deeply than ever before. In this program, we exploited the mouse genome microarray and gene expression profile data analysis to give a new view of radiation related biological events, and testified one of our guesses basically.The first part of the research is the mouse genome microarray experiment and the data analysis of gene time-course expression profile. We used mouse genome microarray to detect the gene experssion of mouse bone marrow after irradated by 6.5Gy γ rays globally at 0 hour, 6hour, 1 week and 2 week. Different expressed genes were selected by statistic analysis and fold changes of gene expression. The different expressed genes were annotated by the bioinformatics databases and applied to pathway localization analysis. By use of hierarchical clustering, K-medoid clustering, intersection analysis and significance analysis of Gene Ontology, we put forward a list of γ radiation related genes. With the strong supports of biological articles data-mining technology, sirt1 gene as one of a series of potential radiation associated genes is highlighted in this research.The second part of this research is about the possible role of sirtl gene in γ radiation. Taking advantage of the patent antisense nucleotide design technology, we find ten antisense nucleotide sites. The effective and specific antisense nucleotide was screened by MTS, RT-PCR and Western Blot. The possible role of this gene in radiation sensitivity and anti-cancer therapy were testified by antisense nucleotide.technology, Western Blot and flow cytometry.
