Research on biochips is advancing rapidly and has yielded several commercial platforms, its significance has increased because of its possible impact on diagnostics and pharmacogenomics. Biochip is a kind of micro-analytic system based on micro-electronics techniques, the main feature of biosensors is the close contact between a biological recognition element and an appropriate transducer that gives a signal on analyte interaction, which could be widely employ to squence DNA, capture the target moleculares or cells. Biochip has overcome the problems of the previous technologies, such as low put-through, poor discrimination, long analysis time and laborious procedures. additionally, it's allow simultaneous detection,reducing the analysis time and facilitating automatic analyzing. With these properties mentioned above, development research of cell-biochip is presented in this paper.The aim of the thesis is to construct the PBSC and find the response mechanic model of cell behavior in PBCS based on theory and experiments. By the technology the application in screening drugs and nano-cell biology were explored. Not only the task had promoted the analysis methods, but had it provided potential experimental proves for clinic medicine, as well as improvement of the nano-cell biology.The methods include combining the cell culture technology with a Piezoelectric Bio-Sensor Chip (PBSC, Homemade) instrument, a real-time analysis system established the PBSC system studying cancer cells behaviors. In order to verify the dynamic process of cell attachment behavior SEM and DTA assay were operated. During the application of PBSC MTT assay were used. At last HD method was used to prepare gold nanoparticles, which were proved by STM and UV photometer. Some achieved progresses have been obtained as follows:1 The analysis model of complexed cell system in PBSC was calculated as follow equations:It concluded the frequency shift was positively relative with impedence of acoustic wave. The impedence of acoustic wave primarily determined the response of PBSC.2 PBSC monitored 3 cell lines, HUVEC, normal liver cell line 7702 and hepatoma cell line HepG2, and compare the difference of response of cell behavior in PBSC.
