Cloning Of Noggin Gene And The Tissue Expression Of Noggin And BMP4 In Xiang Pig From Rongjiang County

Noggin shows more and more important to the regulatation of the growth and development in animal. Noggin is a secreted polypeptide which functions both in early and later development stage. Noggin can make Xenopus embryo restore normal dorsal-ventral body axis that had been artificially ventralized by UV treatment. Noggin induces the nerve tube formation. It binds and inactivates members of the transforming growth factor-beta (TGF-beta) superfamily signaling proteins, such as bone morphogenetic protein-4 (BMP4). Proximal symphalangism (SYM1) and multiple synostoses syndrome (SYNS1) are related to noggin mutations. We amplified the up and down fragments of noggin gene by PCR. The two fragments were cloned into plasmids of pMD18-T-vector. The two recombinant plasmids named pMD18-T-nogup and pMD18-T-nogdown. The recombinant plasmids were recognized by restriction endonuncleases digestion and colony PCR. The whole gene of noggin was made up by the two arrayed fragments. The homology of noggin gene between xiang pig and other species was from 51% to 96%. The forecasted three-dimensional structure of noggin matured peptide of xiang pig demonstrated that the difference of structure compared with human and mouse is due to the the amino acid substitution at 99 and 112 sites.Semi-quantitative RT-PCR was used to investigate the tissue expression of noggin and BMP4 gene in Xiang pig. The results showed that noggin expressed in brain, ovary, and uterus but not in spleen, lung, spinal, liver and kidney.
