Food poisoning case happens frequently in recent years. So food safety is more and more important. HACCP appears to control food hazard.It is abbreviation for Hazard Analysis and Critical Control Point.This system is composed of Hazard Analysis and Critical Control Point. HACCP is a kind of usual technology and analysis tool which may control food hazard.And it is also a kind of important manage system. It may combine with any food processing, which maintain the economic effectiveness of the food safety project.The strains of the genus Staphylococcus were Gram-positive bacteria,of which Staphylococcus aureus was a kind of important pathagenic bacteria with 0.8-1.0 micra diameter,arraying in grape shape. Some strains had capsule or slime layer.Staphylococcus aureus was popularly found in air,water,dust and egesta of human and animal.Some strains of the genus Staphylococcus may produce enterotxin which is primary reason to cause food poisoning.China has become the member of WTO facing opportunity and challenge.In order to make foodstuff enter the worldwide market smoothly, it is necessary to analyze food hazard caused by Staphylococcus strains accurately.This thesis studied the genus Staphylococcus with morphology and molecular biology which would assist the classification of the genus Staphylococcus and develop reagent kit to assay S.aureus in foodstuff.This thesis studied the genus Staphylococcus with morphology and molecular biology which would assist the classification of the genus Staphylococcus and develop reagent kit to assay S.aureus in foodstuff.The thesis studied the genus Staphylococcus in three aspects.1. morphological identification and entertoxin assay of the geneus Staphylococcus In this part strains were collected.Thereafter morphological identification and entertoxin assaying were carried on. Identification ways of the genus Staphylococcus were Gram reaction and APIStaph system test.Assaying ways of entertoxin was enzyme-linked immunosorbent assay (ELISA) .The result showed that fifteen strains were all in the genus Staphylococcus. APIStaph system exsits limitation when color reaction is incomplete.The research showed that several species produce entertoxin.So attention should also be paid to nonpathogenic starins.2.The study on rDNA ITS sequence analysis of the genus StaphylococcusDNA sequences of ten strains were obtained.
