Melon in Xinjiang is famous for its unique quality in domestic and foreign. But in recent years, melon diseases especially downy mildew and fusarium wilt of fungi disease had a negative effect on melon's products and quality. The technique of gene engineering to realize disease resistance molecular breeding is a new approach and will has a broad application in the future. Developing disease resistance genes of melon are our research emphasis in this research on the basis of former work in our laboratory. We selected'Japan annong-2'as material to clone sequences relating to resisting downy mildew (MRGH-J DQ287966) and fusarium wilt(R-Fom-2 DQ287965) by RT-PCR and PCR methods respectively . In addition, we constructed the plant express vector harboring with gene resistance to fusarium wilt(R-Fom-2). Sequence analysis showed: MRGH-J which was 3 kb long encoded 1007 amino acids including a whole open reading frame; R-Fom-2 is long 3307 bp and it encoded 1073 amino acids and its open reading frame is situated between the sites of 1 and 3219 bp. Homology analysis indicated the two genes obtained shared high similarity with the corresponding disease resistance gene published which suggested they maybe be of great important in the process of resistance to downy mildew and fusarium wilt respectively. This research will not only make preparations on studying disease resistance molecular mechanism of melon, but also provide reference values for selecting excellent melon resistance genes by the means of disease resistance molecular breeding in Xinjiang.
