One major drawback in the research of animal mammary gland bioreactors is the low level of foreign gene expression in the milk of transgenic animals. To obtain high and stable expression of foreign gene, the various promoters have been used with constructing expression vector in recent researches. Human lactoferrin, also called lactotransferrin,is a bioactive, versatile protein, and has large potential in nutritional and therapeutic applications. Therefore, production of recombinant human lactoferrin (rhLTF) using animal mammary gland bioreactors was studied widely to satisfy its commercial requirement. The vector was constructed to express rhLTF in animal mammary gland. The rhLTF expression cassette contains the goat beta-casein regulate eliment, hLTF cDNA and enhancer DNA fragment. Enhancer sequence was amplified by PCR. By using transgenic technology (microinjection) and one transgenic mouse (female) was produced. The integration rate of G0 mice was 3.5%. 12 offsprings (10 female, 2 male) were generated, and the...
