Extracellular matrix is important as a scaffold for tissue structure and alsoas an environment regulating cell function in tissues. Matrix Metalloproteinases(MMPs) are a family of zinc metallo-endopeptidases secreted by cells, and areresponsible for much of the turnover of matrix components. The MMPs areinvolved in a wide range of proteolytic events, in normal and pathologicalcircumstances. Normal physiological roles for the MMPs include neuritegrowth, cell migration, bone elongation, wound healing, angiogenesis,ovulation, sperm maturation, uterine involution, menstruation, enamelformation, antigen processing and presentation, mammary gland development,hair follicle development, and embryo implantation. Pathological processesinvolving MMPs include tumor growth and migration, fibrosis, arthritis,glaucoma, lupus scleroderma, cirrhosis, multiple sclerosis, aortic aneurysms,infertility, and many more diseases. It is fair to say that a proteolytic event iskey to a wide range of biological processes, including tissue remodeling andalso modification or release of biological factors. The MMPs areover-expressed during invasion and metastasis and degrade extracellular matrix,lead to invasion and metastasis of tumor. So, one of the important futureprospects is to address the inhibitors that act only on specific MMPs for thetreatment of cancer.We have carried out these works:In order to have the active protein, we found the optimal induce time of therecombinant MMP-17 protein, which is expressed heavily in inclusion body.Six histidines tag make it easy to purify the inclusion protein, because we can purify theprotein by nickel affinity chromatography. Based on the nature of inclusion body, we selectwith subject and fluorescence detection, we confirmed its activity that reached the level toscreen MMPs inhibitors. At last, some subject that may be the MMPs inhibitors had beendetected by fluorescent Microplate Reader. These data will be useful for findingnew drug against cancer.
