Anti-CD3 (OKT3) monoclonal antibody was widely studied and was reported to have the effect of improving success rate in organ transplantation. But the clinical application of OKT3 was highly blocked by its immunogenicity. In this study, anti-CD3 Human/mouse chimeric antibody was constructed by using gene engineering methods in order to decrease its immunogenic properties.To construct the anti-CD3 Human/mouse chimeric antibody, We connected variable region of the heavy chain with the constant region of heavy chain. The anti-CD3 genes,the gene of variable region in heavy chain(Vh)of anti-CD3 genes was synthesized by biology technical service company in ShangHai, and the gene of the constant region of heavy chain(Ch) was cloned by pPICZαIgH vector form our laboratory. Anti-CD3 Human/mouse chimeric antibody could educes biological effect of Fc fragment,such as inducing immune tolerance to reject reaction, prolonging the survival time of the transplant and improving the state of an illness with experimental Autoimmune disease.It was recently proved that low dose anti-CD3 antibody could activate T cell to CD3AK cell, a killer of tumor cell, both in vivo and in vitro.pPICZαIgH, an expression vector of heavy chain, had been precipitated by our laboratory.
