Recently,Studies of stem cellshave suggested the existence of mesenchymal stem cells MSCs in bone marrow,besides of hematopoietic stem cells.MSCs have the ability of self-renewing and differentiation,With different induced conditions,MSCs can differentiate into osteoblasts,chondroblast,adipocytes,neron,and so on.Because it is easier for both acquring,separation,cultivation,proliferation and introduction and expression of exogenous gene,it became the research focus of medicine.The research discuss the way for the isolation and culturation of mouse bone marrow MSCs,and the ability of mouse bone marrow MSCs differentiate into neuron-like cells.The Research compared whole-marrow isolation with density gradient isolation for the purification of mouse mesenchymal stem cells,and investigated the influence of different culture mediums (RPMI-1640 and L-DMEM) and serums (fatal bovine serun and horse serum) in culture mouse mesenchymal stem cells.β-ME and BHA were used for mouse bone MSCs differentiate into neuron-like cells.The result Proved:1 Density gradient isolation can isolate mouse mesenchymal stem cells more purer than whole-marrow isolation,but the groth rate of the cells was slower;After inoculating,cells in the medium were round,different sizes,could not differ the nuclear,but compared with density gradient isolation,whole-marrow isolation contain more cells.The 1st day to the 3th day,the morphology relatively homogeneous,high diopter,spindle cells which was isolate by density gradient isolation were appeared and also some cells of fibroblast morphology,later the cells increased gradually,but the cell isolated by whole-marrow isolation reached logarithmic phase;Approximately the 4th day to the 5th day,the cell isolated by density gradient isolation reached logarithmic phase,but the cells isolated by whole-marrow isolation reached plateau phase;On the 8th day to the 10th day,the cell isolated by density gradient isolation reached plateau phase,and became confluence,but the cells isolated by whole-marrow isolation have confluence already,contact restrain among the cells was present.The purity of mouse mesenchymal stem cells in the 3rd passage and the 4 th passage were more than 98%,it reached confluence on the culture of the 12 th day.the distribution of mouse mesenchymal stem cells appeared whirlpool or parallel line.
