Clone And Expression Of Duck IL-2 Gene

PART ONE CLOING THE IL-2 GENE OF DUCKObjective:To clone and analyse the sequence of IL-2 gene from duck lymphocyte stimulated by PHA.Methods:The total RNA extracted from duck lymphocytes (LC) was used as template. Specific primers based on the published sequence of Il-2 gene of duck were synthesized. IL-2 gene was amplified by RT-PCR using the template and primer. The RT-PCR product after purification was cloned into pQE30 and sequenced, constructing recombinant plasmid namely pQE30-IL2. The sequence acquired was compared with reference sequence of duck IL-2 genes reported in GenBank by software DANMAN.Results:Sequence analysis indicated that the length of cloned sequence is 748bp in this experiment. There was only 2bp difference between the cloned gene and duck IL-2 genes reported in GenBank by homology analysisConclusions:Duck IL-2 gene was cloned successfully.
