Objective: The research is to screen a bacterial strain with ability to form trehalose from glucose, to optimize its trehalose fermentation conditions and to establish a new process of trehalose production.Methods: The trehalose-assimilating strains were obtained by the preliminary plate screen with the medium containing trehalose as the sole carbon resource. Then the flask screen was carried out with a trhalose-forming medium containing glucose as the sole carbon recourse. According to the results of thin layer chromatography analysis of the culture broth, the strain with the highest trehalose yield was chosen. The optimization of the trehalose fermentation conditions of this strain, including the medium composition, the initial pH, the ferment temperature, the aeration and the inoculum of seed culture was studied. After the fermentation under the optimal conditions, trehalose was extracted and purified from the culture broth and applied to the infrared (IR) spectrum analysis. Results: Among all the strains tested, strain Brevibacterium sp. SY361 kept in our lab showed the highest trehalose-productivity. The optimal medium of...
