Apotosis,which also being called Programmed cell death, is a physiological dying form with special morphologic characteristics and biochemical changes of cells. Researches to apoptosis of Bombyx mori. have great theoretical significance to revealing the mechanisms of developmental biology and genic control of absolutely metamorphic insects, as well as important leading theoretical significance to applied researches sericulture science.The main idea of this study is the elementary mechanism of ActD induced BmE apoptosis, and studying the mechanism of silkworm hemolymph inhibiting apoptosis.We analyzed and compared several detecting methods, selected and optimized these methods that adapt to detect the apoptosis of silkworm cell, and ultimately established essential model of silkworm cell apoptosis.The work is helpful to study the apoptosis of silkworm, and it provides the foundation of theory and experiment.The study used morphologic examination and methods of molecular biology and ActD culture, to study the apoptotic character of silkworm cell. And compare several methods such as being stained with Hoechst observed with fluorescence microscope, DNA electrophoresis and Flow cytometry(FCM), to detect the changes of BmE apoptosis. Detect the expression of p35 through NPV inducing apoptosis of BmE, and whether silkworm hemolymph affect the expression of p35 gene.The result were as follws: 1) Changes of BmE cells apoptosis:Morphologic observation of BmE,BmE Transmission electron microscopic appearance of BmE cells after a 12h incubation in Grace medium alone. In normal cells, heterogeneous nuclear chromatin and the marginat ion of condensed chromatin and three or more membrane-bounded apoptotic bodies were obser?ed.
