Mycobacterium tuberculosis (MTB) is the causative bacterium of tuberculosis. With the increase of drug-resistant Mycobacterium tuberculosis , Effective therapy ways hadn't explored. MTB can be dormancy in the host cell, at the same time it can be activated by resuscitation-promoting factor (Rpf). The glyoxylate cycle/shunt of microorganism is very important for survivance in the lack of nutrition. Isocitrate lyase (ICL) is one of the important enzymes of the cycle, and it is required for C.albicans , Salmonella and MTB persistence and virulence. The interaction of ICL with other proteins could further help us understand the relation of MTB and host cell.In this article, yeast two-hybrid technology was used to identify the interaction partner of ICL from human liver cDNA. The protocol was to clone icl. assay of X-Gal, co-transform with cDNA to Mav203, assay of X-Gal, extract yeast plasmid, transform E. coli, co-transform plamid...
