Oxidant stress induced cell, tissue and organ injury is animportant pathologic process, and concerned with many diseases. Now,it has become significant research domain in modern biology. The directresult of biological molecular attacked by ROS is that the electrontransfer function is injured or intermitted. ROS is an special signalmolecular which is necessary to keep body balance. It is reported thatROS can result in many disease, such as heart failure, ischemic heartdisease and Parkinson's disease. In addition, it is also reported thatthere was closer relationship between ROS and senescence.The ascorbate peroxidase(APX)mimics(deuterohemin Histidinepeptide-6,DhHP-6) (3.9×103U·μmol-1) activity is similar to that ofMP-11(4.2×103U·μmol-1). Our study has proved that DhHP-6 has aneffect of on anti-cataract. To investigate the anti-oxidative effect,an experimental model of oxidative injury, which was induced by H2O2(100μmol·L-1), was established by using cultured neonatal rat ventricularmyocytes.The anti-oxidant effects of DhHP-6 were investigated in the modelof oxidative injury by trypan blue exclusion, MTT cell proliferationassay, LDH leakage and the express level of catalase,Hsp70,hemeoxygenaseΙ,and p21 RNA. The results show that DhHP-6 has moreeffective protective effects on anti-oxidative damage. RNA expresslevel of DhHP-6,detected by Real-time PCR is lower than that of H2O2treated group. It proves that DhHP-6 can inhibit cell apoptosis, andhave effective function on signal transduction.The peroxidase activity of MTS~DhHP-6 (2.1x103 U·μmol-1) is similarto that of MP-11(4.2x103 U·μmol-1) .The MTS-DhHP-6 and DhHP-6 are bothThe anti-oxidative effects of MTS-DhHP-6 and DhHP-6 wereinvestigated in the model of oxidative injury by trypan blue dyeexclusion, MTT cell proliferation assay and LDH leakage . The resultsshow that MTS-DhHP-6(5nmol·L-1) has more effective protective effectson anti-oxidative damage than that of DhHP-6 (5nmol·L-1) has.MTS-DhHP-6is an excellent APX mimics and has potential clinical use in theprotection and therapy of the diseases caused by free radical damage.This article founds a definite basis on researching APX mimics.
