Resveratrol is one of a group of compounds called phytoalexins which mainly has the function of antibiosis , resisting oxidization , preventing platelet agglutination , protecting cardio-vascula, resisting cancer and so on. The recent research shows that Res can prolong the lifespan of a kind of vertebrate fish up to 60% than usual, which means this compound will make vertebrates, and possibly humans, live longer .But the content of Res is low in plant under natural environment, so it is difficult and costly to use traditional method to get plentiful Res. RS (resveratrol synthase) is the final enzyme which plays a key role in the way of Res biosynthesis. In this paper, the RS gene was got by molecular biology method and the plant and Pichia pasteoris systems had been established separately.The details show as follows:The RS gene was amplified by PCR from the grape genome DNA and it was cloned into pBS-T Vector. Sequencing result analysed in NCBI database shows :Blastn with nucleotide sequence S63221, the homology is up to 98%;with DQ459351.1 is up to 99%. Blastx with amino acid sequence ABE68894.1, is up to 98%.The RS gene of grape contains1179 base pair and encodes 392 amino acid,which proved the pBS-T Vector had been successfully constructed.Moreover, plant expression vector pBI 121/RS had been constructed by enzyme digestion and ligation which contained CaMV35S promoter and NOS terminator ,and the recombinant plasmid was transfered into agrobacterium LBA4404 by freeze and thaw method.Then the tobacco leaf disc was transformed with LBA4404/RS and the regeneration system had already been established .HPLC analysis showed Res had been produced by the transgenic tomato.The recombinant vector pPIC3.5K/RS had been successfully constructed whichcontains the AOXl promoter and HIS4 gene ,and the target gene was integrated into Pichia pastoris genome via homologous by electroporation.The fermentation experimentation and SDS-PAGE analysis showed that the target gene had already expressed.
