Abstract: To our knowledge,it is the first time to analyze Hesl functioning indifferentiation of hepatic stem cells. Hepatocyte specific markers, Glutamine Synthesase and Biliary Glycoprotain, were downregulated. In contrast, Specific markers of biliary epithelium cells, such as CK19 and GGT were upregulated. HNF6 and HNF1 β were also detected to be upregulated, which play important roles in generation of biliary epithelium cells. Our data certificated that the expression of Hesl can induce the differentiation of hepatic stem cells to biliary epithelium cells initially. However, the precise mechanism of how Hesl determines the biliary cell fate is not known yet, and further study is necessary to clearity the function of Hesl during hepatic stem cells' specification. It will be invaluable for the research of bile duct and the therapy for biliary diseases.
