In this paper,we used spiral algae ,marine chlorella and limnetic chlorellaas experimental material. Spiral algae and marine chlorella cultivated by metal ionaddition in the culture medium. we use Ca-alginate to immobilize microalga cells andrestore them in the fridge at 4℃ for a long time.First the marine chlorella is cultivated in the culture medium with 6.5 in PH, 24 ℃and 10 hours: 14 hours light circulation. And we can get bright color and strong vitalityof marine chlorella.we add Mn2+,Cu2+,Cd2+,Zn2+,Co2+ of different concentration inthe culture medium to study the toxicity function of the heavy metals ion towards themarine chlorella ,and to analyze the tolerant ability of marine chlorella and the deathlyconcentration of the heavy metal . The result shows that the inhibition of marinechlorella will be stronger if the concentration of heavy mental ion is higher. Theendurance of marine chlorella is different to Mn2+,Cu2+,Cd2+,Zn2+,Co2+, to the Co2+,Mn2+,Zn2+ it is stronger and to the Cu2+,Cd2+ more sensitive, the death concentration ofthe Co2+ to marine chlorella is 2*10-4 mol/L , the death concentration of the Mn2+ tomarine chlorella is 1.5*10-4 mol/L, the death concentration of the Zn2+ to marinechlorella is 2.5*10-4 mol/L, the death concentration of the Cu2+ to marine chlorella is2.5*10-4 mol/L 5*10-5 mol/L, the death concentration of the Cd2+ to marine chlorella is2.5*10-4 mol/L.Spiral algae is cultivated in the culture medium named Zarrouk and researchingthe changeful condition of the value of OD and biological quantity in normal growingcondition and abnormal condition that has been added glucose. The result shows thatthe spiral algae can grow well in the culture medium added glucose,but it is easyinfenct by bacterium.we add Mn2+,Co2+,Cd2+of different concentration in the culturemedium to study the toxicity function of the heavy metals ion towards the spiral algae.The result shows that spiral algae has more strong endurance than marine chlorella byMn2+,Cu2+,Cd2+ addition in the culture medium. The death concentration of the Co2+ tospiral algae is 1.25*10-4 mol/L , The death concentration of the Mn2+ to spiral algae is3.9*10-4 mol/L, The death concentration of the Cd2+ to spiral algae is 2*10-4 mol/L.Last we carried on alginate immobilization experiment with different measuresrespectively. The conclusion of optional condition is that we used 2.5ml injector, 4%Na-alginate , 0.1 mol/L CaCl2.The green microalgae was cultivated and entrapped intoalginate-beads for long-term storage in absolute darkness, at 4oC without the liquidmedium. The entrapped marine chlorellas and limnetic chlorellas were alive andmaintained their physiological activities after one year of storage. spiral algae canstorage for four months. Immobilizated microalgae is a good method for microalgaestorage.
