Ocean is the cradle of life. With the developing of technology and the more comprehensive of people's acquaintance to the nature, we have made progress in halobios studying from microscopical to microcosmic, from local action to whole action, from correlated groups have direct economic effect to ecological communities have systemic effects. However, until recent years, people's understanding on marine virus is still much behind it's real fact.Diversity viruses are abundance in the sea. They play a very important role in marine ecosystem. Diversiform marine viruses probably infect all organisms. Marine viruses influence many biogeochemical and ecological process, they are the largest cycle in biogeochemical cycle. The ecological characters of viruses have a significant impact on the occurrence and spread of marine organism diseases. It is known to all that viral disease has led to great hurt and damnification to the world's aquiculture industry. It is the main problem that should be soloved in aquiculture industry developing.So, the coming research fields in marine virus include the primary survey of the distribution, abundance, resource library and gene library of virus isolates from different oceanic zone, will give a basis to ocean ecosystem dynamics and marine organism disease research, and will have great meaning to the innovation of ocean ecosystem dynamics, prevention and cure viral disease in aquiculture industry, and quickly detect and identify virus in seawater.To extract and purify viruses from seawater, we have tried different way to concentrate and isolate viruses from seawater, such as absorb and wash off, chemical flocculate, and so on. Finally, we isolated some free viruses from Qingdao coastal waters successfully by using AlCl3 sorbent. Transmission electron microscopy observation shows that, there were free viruses in seawater, and their type was various. These viruses' shape include globosity with and without thread, bacilliform virus,bacteriophages, and so on. Their size were about 40~400nm long. We set up a simple and effective extracting method to isolate viruses by concentrate a large volume of seawater at the same time.We have tried to construct a DNA library of marine virus, the DNA fragments we used were ruptured by physical strength and cleaved with EcoR I and BamH I, but we have not got a clone have inset fragments. Quantity analysis shows that the concentration of the virus is too low, the type of the mixed virus DNA is unknown. Effected selecting method should be set up to identify different virus.To have a concept of the count and distributing of the virus in seawater, we use fluorescent microscope counting stained with SYBR Green I to research the free virus' quantity in seawater in Qingdao. Quantity analyzing by the photo analyze software shows that, virus' concentration in seawater have great difference in different time and different place. At the same time, it shows that fluorescent microscope counting stained with SYBR Green I is a simple nicety and high sensitive method. It can be used in the virus' dynamic quantity research in the water.
