ObjectiveSleep is one of the basic characterastics of life,and it is one of the essential necessaries to maintain life.Sleep Deprivation (SD) is the state that the amount of sleep is insufficient because of different causes.SD has great effect on body. This research focus on the effects of SD on myocardium and its possible mechanisms,using flower pot technique to establish the experimental model of SD.Method1 Subject:Sprague-dawley(S-D)rat,weighing about 240~260g,half male and half femal.S-D Rats were radomly divided into five groups:Normal Cage group,Tank Cage group,SD 2d group,SD 4d group and SD 6d group.2 Experimental model of SD.Using flower pot technique to establish experimental model of SD.The glass box of 30cm X 30cm X 30cm was made,with a platform (6.3cm in diameter,8cm in heighten the center of the box. 14cm above the platform,a lid proper to the boxwas placed on which were water and food supplied for the rat. When doing experiment,water was put into the boxjust lcm lower than the platform, and the temperature of the water was kept at about 20 centi-degree. The condition of the tank group was identical to the SD group except that the former had a larger platform which was 18cm in diameter.2 Observation of general state of healthDuring the courese of SD, observe the general state of health, the emotion changes and the response to stimulus.3 Recording of electrocardiogram(ECG)Under anesthesia state,using needle electrode and BL-410 data acquisition and analysis system for life science to record the standard II limb ECG.4 Miocardium morphology invastigationChose the cardiac apex part of cardiac muscle,sent for the electron microscope preparatio.Part of the left ventricular cardiac mussle was cut off ,put into 10% formaldehyde ,using paraffin buried for HE staining.5 MDA content and SOD activity of myocardial mitochondriaExtract myocardial mitochondria by differential centrifugation.Using Coomassie Brilliant Blue to detect the protein concentration of the mitochondrial suspention.And detect the MDA content and SOD activity according to the correspond detection kit provided by the Nanjing Jian Cheng Bio-company.6 Statistical analysis:All data were expressed in form of x ±s.Using one-way ANOVA tocampare the differece between groups.Dunnett-t test was chosen to make sure if there was difference between percific group.Significance level was chosen ata =0.05 and all tests were two-sided. Results1 Changs of general state of healthAfter SD,The rats were depressed , emaciated ,the fur was bloom, their excitability was higher and the rats responsed more fiercely to stimulation.At the sixth day of SD,rats escaped from general stimulation such as petting,looking like suffering to the stimulation.2 Effects of SD on ECGThe heart rate of the SD groups and the tank group was higher compared with that of the nomal cage group (p<0.05) .S-T segements of the SD groups were higher to that of the normal cage group.And there were some S-T segments mix with T wave of ECG..P-R interval and Q-T interval were prolonged in SD groups .3 Eeffects of SD on the myocardium morphologyThe morphology changes of the SD group under normal light microscope of HE stain were as flows: there were edema ^ congestion > hemorrhage in myocardium matrix. The memebrane of the cardiac muscle were blured,and signs of myocardium dissolution and cadiocyte membrane hyperplasia could be seen in some part of the cardiac tissue The morphology changes under electron microscope were:the chromosome of myocardium was dissolved gently, the endoplasmic reticulum was expanding and there was evidence of over loaded of Ca +;the stucture of mitochondria was blured;the vessel in the matrix could be seen accumulated with thrombocyte. Myelin bodies could be easily seen in myocardium cells.The morphology changes of SD suggested that there was severely damage on myocardium under SD statement.4 Effects of SD on MDA content of myocardium mitochondria...
