Classical Swine Fever is a harmful disease to swine husbandry. Classical Swine Fever virus (CSFV) is its cause. And its main protective antigen is glycoprotein E2. Animals can prevent from virus attack by using protein E2 vaccine prepared from eukaryotic expression system.CSFV E2 gene drived by tuber specific expressed promoter -Patatin is inserted into plant nucleus transform vector-pBI121 to construct specific expressed vector named as pBIPE2, which contains: antibiotic gene (Kan), Patatin promoter and foreign target gene(CSFV glycoprotein E2 gene ). The vector pBIPE2 was transferred to Agrobacterium LBA4404 and the Agrobacterium LBA4404 containing E2 gene was cocultured with the potato leaf. Callus and shoot were induced on the selective medium and regeneration system adventitious shoots was gained, the regenerated plants on selective medium were used for following analysis, PCR analysis and Southern blot analysis show that E2 gene has been integrated into genome of potato.
