Through examining the DNA damage and the Ca2+ density of rat hepatocyte, which was irradiated by three kinds of different wave bands of UV, the relation between the DNA damage level and the irradiative dosage of UV was determined. [Ca2+]j during the damage was detected. Then the mechanism of UV damage and repair was revealed.Besides the types and the reasons of the DNA damage on biology cell, the various methods used to determine the damage were introduced, and the mechanism of Ca2+ signals action in the process of the DNA damage was analyzed. Using single-cell gel electrophoresis, the DNA damage was studied when the small rat liver cell was irradiated by different wave bands and different dosage of UV. [Ca2+]; was tested by ratio fluorescence imaging system. Analyzing the data of the tests, the graph of relationship between the degree of DNA damage and the quantity of different UV's wavelength was drawn out. The results were shown as follows:(1) Whether the UV can damage the DNA lies on the UV's wavelength(or frequency);(2) The irradiated energy of UV is in direct proportion to the degree of the DNA damage;(3) There is the feature of high energy flat and low energy threshold value in the graph;(4) [Ca2+]i will rise during the damage. The amplitude and the frequency will vary with time.
