Enhalus acoroides , the monocotyledon hydrocharitaceae seagrass, are rich in the South China Sea. The constituents of Enhalus acoroides had been studied by various seperation methods and their structures were determined by chemical methods and spectral methods. Moreover, the crude extracts, polysaccorides and some pure compounds were experimented the antibacterial, antivirus (CVB3, HSV-1) and the activate activity of insulin secretion.Ten compounds were isolated from the sea-grass Enhalus acoroides by various chromatographic methods such as silicon chromatographic, Sephadex LH-20, TLC and HPLC along with the traditional methods such as extraction and crystal. There are 8 compounds structures had been determined, they are luteolin(EII-1), Chrysoeriol(EII-2), 2-(2,5-Dihydroxy-phenyl)- chroman-3,5,7-triol(EII-3),Stigmasta-3- 3 -D-glucopyranosyl(EII-4), 4-Hydroxy -benzoic acid(EII-5), 1-methyl-3-piperidinemethanol(EII-6), Luteolin-7-O-D-glucur onopyranoside(EIII-1), Chrysoeriol-7-O-D-glucuronopyranoside(EIII-2). All these compounds are isolated from Enhalus Rich for the first time of them.The polysaccharides of Enhalus acoroides were extracted by hot water. Savag method was used to remove the protein and the content of the protein was measured by UV spectral method. The contents of total polysaccharides, sulfate and monosacchrides were analyzed by the methods of anthranone-sulfuric acid, BaCl2 tuibidimetry and GC-MS.The minimal inhibitory concentration (MIC) of the rude extaction, polysaccharides and several pure compounds of Enhalus acoroides were investigated. The polar fraction of Enhalus acoroides was found to have good inhibition activities against the Gram-positive bacteria. The MIC what the water extact of Staphylococcus aureus is 0.156 mg · mL-1.The inhibitory activities against CVB3 and HSV-1 were studied by cell culture in vitro. The cell cytotoxicities of the samples were evaluated by MTT method, theinhibitory effects were evaluated by cytopathic effect (CPE) reduction method. The result showed that the water extract (Ew) and the polysaccharides (Ep) had potent CVB3-inhibitory and HSV-1 inhibitory efficacy and low cytotoxicity. The IC50 of Ep against CVB3 and HSV-1 are 3.908 μ g · mL-1 and 15.63 μ g · mL-1 , respectively and the IC50 of Ew against CVB3 and HSV-1 are both 15.63 μg · mL-1.The activate insulin secretion activity of Ep was studied in vitro. Although the activate activity was unobvious, Ep was found to have low cytotoxicity towards the pancreas and was expected to carry out the experiment to cure the pancreas which were hurt by high concentration sugars.
