Cellulose is a kind of reproducible resource of great output, it takes about a half of the hundred million biomaterial making by photosynthesis. It is of great importance to transfer these resource to energy ,food, and so on. The main obstacle of using cellulose resource is the low celiulase activity. On one hand , the it is because the structure of the celiulase itself, on the other hand, we need deeper research of celiulase binding, cacatalyse activity.In this study, first, we express the Trichoderma reesei 's Endocellulase in Pichia pastoris. After this, we contrast the expression ability of the recombinant which has the different gene copy of EGâ…¢.The structure gene of EGâ…¢ were obtained by PCR. This gene was inserted between the pPIC9K's AOX1 promoter and the TT point. Then, the reform plasmid pPIC9K-EGâ…¢ was transferred to P. pastoris GS115 yeast. And was expressed by this yeast. All the recombinant yeast strains secreted EGâ…¢ protein by the abduction of methanol.Second, we launch a study of the Kinetics of cellulose hydrolyze by EGâ…¢. In this section, we form a Kinetics model based on the Theory of Protein Adsoped on the solid surface. Then, we try to use the experiment data to calculate out the parameter in this Kinetics model.
