Nitrile Hydratase (EC4.2.1.84, NHase) is a kind of enzyme from microorganism, it can catalyse a series of nitrile hydrolyze to acylamideo In this paper ,we studied nitrile hydratase from Rhodococcus rhodochrous Jl.We studied how the enzyme come from ,the enzymologic characters and its application.All of our studay of nitrile hydratase are comprised in the list. Its maximum activity could achieve 6150u/mL when culture mediums, temperature, and pH were adjusted. Given the enzymologic characters ,the temperature of 30D and the neuter system are benefit for its activity and stability. The orthogonal-experiment that test of the reaction time ,the saturation of substrate and the enzyme had found that the significant factor was the substrate(acrylonitril) that the best saturation is 5%. Over this saturation, acrylonitril would kill the enzyme .In the best conditions, the invert rate could achieve 80% in one minute.The fixation of the cell can't improve the stability of the enzyme.Then,we purified and characterized the nitrile hydratase of Rhodococcus rhodochrous J1. The enzyme has a molecular mass of about 505kD,and consists of two different subunits(a,23.1kD, B ,27.4kD) .The purified enzyme has less stability .We discovered that consecutive reaction by free cell is the best method for the production of acylamide.
