The dissertation made research about the deep-sea Streptomyces, included collection of samples, the separations and conservations of strains, the screen of biological activities, identifications of strains, optimization of culture medium, distillation of metabolites, and purification of natural products. In order to make use of its biological activity, preliminary explorations are made of its metabolites.The samples from which eight actinomycetes are separated were collected in Northwestern Pacific Ocean. In virtue of microbial model, included Sarcina, Staphylococcus aureus, Escherichia coli PCI 219, Pseudomonas aeruginosa, Cryptococcus meningitis, Candida albicans, Streptomyces viridochromogenes, Mucor miehei, Streptomyces sp. DS003 can inhibit more microbes more intensively. In order to its biological characteristics and value in use, we identified its model characteristics, physiological and biochemical characteristics, 16S RNA, and enzymatic activities.For further increase exploitation in biological activities of the germ, substrate is reasonably optimized in the research. First of all, we choose better substrate from six that comes from M&P institute in Germany .At the base of it, we formulate mathematical model of makeup of substrate by the way of Uniform Design. After validating, inhibitory actions aiming at model have been increased, such as 44% for Escherichia coli PCI 219, 44% for Pseudomonas aeruginosa 21% for Staphylococcus aureus, 40% for Sarcina, 21% for Cryptococcus meningitis, 20% for Candida albicans. The output of metabolite also increases by 33.1 %.Its leavening was made primary purifications of extraction. Wiping off unctuous substances, extraction is made primary purifications by TLC, PTLC, Pharmadex LH-20, and divided into several ingredients, ultimately we found that two of them have better biological activities but ingredients are complicated, so they still need further purifications.
