In the present thesis, effects of nutritional and environmental factors on the growth of Cladosiphon okamuranus and the sensitivity of Cladosiphon okamuranus to six antibiotics were investigated primarily.The investigation showed that Cladosiphon okamuranus could acclimate high temperature, but could not acclimate high temperature above 31; temperature between 19 and27 was its apt temperature extent.Cladosiphon okamuranus has a wide range of photosynthetic adaptability, under illumination of it could grow normally, furthermore the growth rate of Cladosiphon okamuranus increased as illumination rose from 34 mol m-2 s-1-70mol m-2 s-1. Cladosiphon okamuranus could grow rapid at photoperiod from 12L/12D to 14L/10D.Under salinity from 25 to 41, the growth rate of Cladosiphon okamuranus had little change with culture time extending; Cladosiphon okamuranus got the most growth rate at salinity 33.From pH8.0 to pH8.5, Cladosiphon okamuranus got similar growth rate; Cladosiphon okamuranus grew more rapidly at pH8.5; at pH6, the growth of Cladosiphon okamuranus stopped; at pH7.5, pH9.0, pH10, the growth of Cladosiphon okamuranus was inhibited remarkably.When NH4NO3 was used as nitrogen resource, Cladosiphon okamuranus grew more rapidly and maintained longer at logarithmic period than used NH4Cl and NaNO3 as nitrogen resource of the same quantum; the apt range of N resource was from 0.3m mol/L to 0.6 m mol/L, and the middle-high concentration (more than 2m mol/L) of NH4NO3 inhibited the growth of Cladosiphon okamuranus to some degree.In case of different concentration of phosphorus, Cladosiphon okamuranuspossessed well-nigh same growth rate.Cladosiphon okamuranus cultured with different culture medium got different growth rate, maximal growth rate appears when Cladosiphon okamuranus was cultured with F medium. Meanwhile 2 ind/mm2-4 ind/mm2 consistencies was the best culture method in fed-batch cultivation.VitaminBi, VitaminBn and Biotin could increase the growth rate when increased to some content, but more concentration had no negative influence on the growth of Cladosiphon okamuranus.Cladosiphon okamuranus cultured with l/2Provasoli, l/2Provasoli +complex Vitamin, Nori medium got similar growth rate, Miquel medium has a second effect.The sensitivity of Cladosiphon okamuranus to six antibiotics -Chloromycetin, Neomycin, Kanamycin, Ampicillin, Streptomcin, Erythromycin was studied which would be the selectable agent for transformation. The experiment results indicated that Kanamycin(10 mg/L-600 mg/L), Neomycin(l mg/L-100 mg/L) and Streptomcin (1 mg/L-100 mg/L) could not affect the growth of Cladosiphon okamuranus evidently; Ampicillin and Chloromycetin accelerated the growth of Cladosiphon okamuranus at low concentration (less than 10 mg/L and 0.1 mg/L respectively), but obviously restrained the growth of Cladosiphon okamuranus when their concentration increased; Erythromycin could inhibit the growth of Cladosiphon okamuranus remarkably at quite low concentration (1 mg/L).This paper presents the preliminary results on the identification of Cladosiphon okamuranus diploid by means of flow cytometry (FCM). FACScan FCM measured samples. The experimental results show that FACScan FCM can be used to distinguish Cladosiphon okamuranus diploid by measuring emission fluorescence from DNA which has been dyed by PI pigmentation for a long time. Chlorophyll was extracted by methyl alcohol. The optimization of analysis is that Cladosiphon okamuranus is diploid.
