Effect of different Se(IV)-intimidation on the growth of S. maxima and S. platensis at different growth phases, bio-organization and metabolism of Se(IV) were investigated in this study. Total Se content in five biological samples was determined by diaminonaphthalene (DAN) fluorometry. Some interesting results were observed.First of all, Se(IV) concentration increased with adding Se(IV) from the 1st day to the 5th day, gradually. The cumulated Se(IV) concentration on the 7th day was 1000mg-L-1(Se(IV)-intimidation I~V). The growth of Spirulina, the absorption and bio-organization of Se were also discussed. The results indicated that Se(IV)-intimidation I~IV showed no significant effect on the growth of Spirulina,while Se(IV)-intimidation V showed obvious promotion. The total Se content and bio-organization ratio of Se increased from I to V. The results were explained by Se(IV)-intimidation intensity at the first time.Secondly, Se(IV)-intimidation on 5. maxima and S. platensis on 5th and 6th day was implemented, with cumulated Se(IV) concentration of 600, 800 and 1000mg-L-1. The organic Se content was 580.83 , 685.94, 771.65 g g-1(DW) in S. maxima, while 461.91, 616.06, 1376.20 g-g-1 (DW) in S. platensis. The metabolites of Spirulina under different Se(IV)-intimidation were distributed as follows: Se-containing organic molecules in Spirulina; Se-containing organic molecules in algae culture; Se-containing organic molecules volatiled into the air.Thirdly, Se content in five biological samples including Spirulina, tea leaf, medlar, yeast and mythic fungus was determined by diaminonaphthalene (DAN) fluorometry. The detected limitation was 0.223 10-3 g/mL, with liner range of 0.223 10-3~ 1.0 g/mL, recovery ratio of 94.8-104.2%, and average relative standard deviation (RSD) of 4.6%.
