Sinorhizobium meliloti Strain 1021 is able to transform ornthine into proline by means of an ornthine cyclodeaminase and .therefore, has the ability to use either of these amino acids as its sole nitrogen and carbon source .10 recombinant plasmids were constructed via pK18mob,then mobilized into Rm1021 by triconjugation or electroporation, through homologous recombination caused by a single crossover event, selected for Km and Sm resistance, them 8 Rm1021 mutant derivative strains were obtained.Both nonpolar mutant and polar mutant of SMC01053 are able to use either ornthine or Proline as sole carbon and nitrogen source. In TY medium , they are similar to Rm1021, show a little delay in proline medium. Both nonpolar mutant and polar mutant Of SMC01054,SMC02124,SMC02123,SMC02122 can grow in proline medium, but, they can not catabolize ornthine.The pGXHN200 of HN01 genomic library containing ORF homologous to SMC02123, SMC02122 was transfered into SMC02123F/Rm1021 and SMC02122F/ Rm1021 by electroporation or triconjugation with the helper pRk2073,and the mutant strains rSMC02123F / Rm1021, rSMC02122F / Rml021 were achived Inoculating rSMC02123F / Rml021, rSMC02122F / Rml021 into ornthine medium, we found That, they restored the ability to metabolize ornthine.We concluded that SMC01054, SMC02124, SMC02123, SMC02122 are related to metabolism of ornthine.but, SMC01053 has no relation to metabolism of ornthine...
