Bombyx mori prothoracicostaic peptide (Bom-PTSP) is a recently discovered important neuropeptide of insect, produced by brain neurosecretory cells of Bombyx mori. It inhibits the stimulation of prothoracicotropic hormone on ecdysteroidogenesis by prothoracic glands in vivo and in vitro. Prothoracicotropic hormone can activate PG when PG is not active, but this stimulation can almost be inhibited when PTSP in present. Ecydsteroid is one of most important hormones in insect. It controls the growth, development, ecdysteroidogenesis, metabolism and so on with another most important hormone-juvenile hormone.In the present dissertaion, PTSP solution conformation and its potential applications were studied.1. Conformation analy/e of PTSP in DMSO-d6 with NMR spectraIn the dissertaion, PTSP was obtained by sollid phase method. Its solution in DMSO-d6 was examined by 2D NMR technique. The COSY, NOESY and 13C HMQC-COSY spectra were applied for the analysis of the conformation. All resonance assignments of 5H NMR were made with the two-dimensional correlation spectroscopy. The problem of sequential resonance assignments was solved by the analysis of NOESY spectroscopy. All 1H and 13C chemical shifts of PTSP were identified. The chemical shifts of 1H and NOEs stronly suggest that PTSP exists mainly as a flexible random coil in2. Studies of PTSP as a potential novel pest control agent and other latent applicationsProthoracicostaic peptide (PTSP) inhibits the biosynthesis of ecdysone when prothoracicgland(PG) of the silkworm was active. It cooperates with PTTH to regulate and control ecsysteroidogenesis on PG of insect. So it is a potential pestcontrol agent. In the experiment of PTSP feeding, PTSP may possibly be aborbed by silkworm and cause the lost of weight and even the death.In proper future research, it will be applied in pest insect bioactivtiy tests. With bioengineering technology, PTSP gene may be transducted into crops and specially infect virus and produce toxicity or add toxicity of the virus to the special species. It can be used in pest management.
