Phenylalanine ammonia-lyase (PAL) can be induced by many kinds of factors including environment , plant hormone and so on. It has effect on the self-protection of plants. So they have a profound role in plant growth and development. ACBF is a transcription factor that combined with the AC-rich region of the promoter region. It has been cloned at Arabidopsis thaliana, Nicotiana tabacum and so on.A pair of degenerate primers were designed in the conserved domain which based on the alignment of ACBF gene family in tobacco and arabidopsis. A 239 bp fragment was amplified by RT-PCR (Reverse Transcription Polymerase Chain Reaction), which was used as a probe for screening tomato fruit (pink stage) cDNA library. One positive clone containing entire coding region were isolated, which was identified as a new member of ACBF family by BLAST Server, and named as LeACBF. The LeACBF cDNA is 1503 bp and the predicted translation product is 428 amino acids. LeACBF was homologous with that gene of tobacco with 83% similarity at the amino acid level and, with 79% similarity at the amino acid level. The cDNA sequence of LeACBF was deposited in GenBank and the GenBank Accession Number is AY506544.Northern blot hybridization showed that LeACBF has well-distributed expression in different tissues of wild tomato (pink stage).Treatment the seedling with UV made the enzyme activity of the PAL increased gradually and then decreased. Northern blot hybridization showed that LeACBF expression had the same tendency with the enzyme activity. And it showed that the transcription factor ACBF maybe have some relation with the enzyme activity of PAL.
