The venom of Chinese bird spider Ornithoctonus hainana has many toxic components. The venom was distilled from spider ornithoctonus hainana by tube. The content of proteins in venom is 79.6%.The LD50 values of venom is 0.042mg/g.A new insecticidal toxin named hainana toxin a(HNTXa) was isolated from the venom of the spider Ornithoctonus hainana by reverse phase chromatography(RPC) and cation exchange chromatography. The map of SDS-PAGE of this toxin indicates that it's molecular weight is less than 14400Da. The map of IEF-PAGE indicates it's isoelectric point is between 8.15 and 8.45. HNTXa is toxic to bollworm(Lepidoptera) and cricket(Homoptera).The LD50 values of this toxin is 0.058mg/g.Ten amino acids of N end were determined. It showed two results. One is that this toxin contains two proteins, the molecular weight of these proteins are similar to each other. The other result is that HNTXa is a protein which has two peptide chains.Reverse phase chromatography(RPC) has many advantages, such as speediness, high effect etc.The technology of reverse phase chromatography is used in purification of spider venom mostly.
