Dissociation, Culture and Identification of Neural Stem Cells from SVZ of Embryonic RatObjective: To gain neural stem cells which have been considered as an excellent source forstudying the proliferation and differentiation of these neurospheres.Methods: The neural stem cells from the subventricular zone of rat embryonic day 14 (E14)were cultured in the serum-free medium consisting of DMEM/F12 (1:1), B27 supplement,bFGF. Identification was carried out using Nestin antibodies for undifferentiated cells. Inorder to determine the phenotype of the differentiated cells, MAP2, GFAP, CNP, distinctivemarker for neuron, astrocytes and oligodendrocytes were used.Results: (1) In primary cultures and subcultures of embryonic day 14 of rats expressed nestin,a marker for neuroepithelial precursors. (2) It shows that the neural stem cell sustained toproliferate in the present of bFGF in vitro. (3) The bFGF -responsive cell was able to generatethe three major phenotypes of the mammalian CNS- neurons, astrocytes, andoligodendrocytes in the serum medium.Conclusion: These embryonic precursor cells exhibit properties and satisfy criteriarepresentative of stem cells in vitro.
