Rat liver bears intensive capability of regeneration after hepatectomy inducement, which involves core problems of life science during the biology course, such as reconstructing and regeneration of mammals tissues and organs, cellular multiplication, differentiation and dedifferentiation, stress response and regulation of physiology and biochemistry. Expression of abundant genes is altered in quantity and quality. To find out their differences and their internal relationship and their real relationship with liver regeneration is the base of explicating molecular mechanism of liver regeneration. In the study, we adopted short interval successive partial hepatectomy (SISPH) 0-4-36-36-36h established by Cun-shuan Xu et al. as experiment model. We used 76h and 122h regenerated liver as experiment material to extract their mRNA and reverse transcript it to cDNA, recurring to suppression subtractive hybridization (SSH) to construct efficient forward subtracted cDNA library, which included up-regulated expressed ESTs of 0-4-3 6-3 6-3 6h SISPH. We picked out 53 ESTs to make sequence analysis and homology blast and found that 28 ESTs was 100% homologous and 25 ESTs was different but high homologous to reported genes. Among the genes, there were genes directly related to liver regeneration: fetuin, cathepsin; close related to liver function: cytoplamic aspartate aminotranferase, gutathion sulfur transferase; related to substance and energy metabolism: ATP synthetase, ribosomal protein, and related to stress response: haptoglobin, transferrin. Cloning these ESTs established the base of making further study the function of related genes in liver regeneration.
