Dunalliena salina, eukaryotic unicellular green alga, is able to proliferate under saline conditions ranging from 0.1 to 5.5 mol/L NaCl. We have cloned and sequenced large-scale cDNAs of Dunalliena salina to gain full-length novel genes. As so far, about 10,000 cDNA clones have been tested, including 2,282 unigenes. Four patents have been applied in the field of transgenic plant to the State Intellectual Property Office of P.R.China.We assorted novel genes according to different standards and predicted their functions, using two high throughput technologies-bioinformatics and microarray assay. Furthermore, we research the function of some novel genes. Our data suggested that functional genomics could be largely facilitate by microarray assay.Bioinformatics is another efficient technology of large-scale function prediction. In this case, we analyzed 2,282 genes with the help of bioinformatics tools, including similarity analysis on both DNA and protein level, domain structure and motif analysis. Our results showed that bioinformatics analysis could suggest possible functions of many different novel genes. For example, we predicted that gene DSG10 could encode nitrate transporter, which was confirmed by our further experiments.In a word, we obtained large-scale cDNA clones and sequences of Dunalliena salina, including thousands of novel genes, and established systemic technologies for functional genomics of that kind of alga.
