To provide evidence for illustrating the molecular mechanism of pancreas development, proteins were extracted from rat pancreas tissue at various stages, separated by high resolution two-dimensional electrophoresis (2-DE), and the good 2DE pattern including resolution and reproducibility was obtained. After silver staining ,the 2DE image analysis by Imagemaster 2DE software had detected 800+50 spots in E18.5 pancreas map and 1000+50 spots in adult pancreas map, of which 8 spots increased and 20 spots decreased in quantity. Furthermore, 4 spots were missing and 14 new emerged in adult rat pancreas compared with E18.5 rat pancreas. For protein identification, proteins spots of interest on the gels stained with colloidal Coomassie Brilliant Blue G-250 were excised, digested in-gel with trypsin, and analyzed by peptide mass fingerprinting(PMF) with matrix assisted laser desorption/ionization-mass spectrometry (MALDI-MS). ERP60, trypsinogen, proelastase, lipaseandso on were identified. This will enable us to present an overview of the proteinsexpressed in rat pancreas tissues and lays the basis for subsequent comparative proteome analysis studies with pancreas development.
