Since Flemming discovered penicillin,more than 350 kinds of antibiotics have been used in clinic, which makes many epidemic diseases controlled.However,people must find new drug continuously because of the wide use of antibiotics and the emergence of bacterial drug resistance.Biological resources in land once were the main source of antibiotics.However, with the research going on, it is becoming more and more difficult to find new antibiotics from biological resources in land. Sea and ocean has particular environments,so marine biology has more biological diversity than terrestrial biology. Marine biology is becoming one of the hotspots of the new drug research in the world.1,451 marine microorganic strains were isolated from 127 selected marine environment specimens in 19 locations of Hainan Island and surrounding areas,including 656 bacterial strains,419 actinomycetic strains and 376 fungal strains. 1,451 strains fermentation products were tested by agar difussion method to screen the antibacterial bioactive substances.As a result, 13 microorganic metabolites have prominent antibacterial bioactivity towards Staphalococcus aureus..HSL I -306 was a actinomycete isolated from the mire of mangrove in Hainan,agar diffusion method showed the metabolities of the strain had prominent bioactivity inhibiting Staphalococcus aureus, and the antibacterial bioactivity can not be lost after screening again. The taxonomic investigation on nutritional and morphological characters display the HSL I -306 strain is Spirillospora sp..The fermentation condition and gradients of the medium have been studied,the results showed the optimum fermentation condition were 48 hr of inoculation age, 96hr of the fermentation time and an initial pH of 6.5-7.2,on that condition. Orthogonal test experiment showed the best culture condition and the gradients of fermentation medium were pH6.5 of the fermentation medium, 300rpm of the oscillation frequency, 28 ℃ of the culture temperature and the gradients of the fermentation mediumincluding 2% of Sucrose, 1.5% of Soybean Powder, 0.15% of Yeast Extraction, 0.05% of CaCOs and 0.20% of Glycerol. Under that condition the strain can produce the most concentration of the bioactive compounds.The physical and chemical properties of the actibacterial compounds have been studied,the results showed the bioactive substance has heat stable,acidity and weakly alkali stable.It can be dissolved in acid water, Chloroform solvent,can not be dissolved in Ethyl Acetate, Petroleum ether,alkali water.The result of Thinlayer Chromatography showed the bioactive compounds can give off blue and green fluorescence in the UV light.The Paper Chromatography demonstrate the bioactive substance may be a new construct compounds.The bioactive strain's fermentation product was isolated and purified primarily using methods of solvent extraction,acid-alkali extraction,AB-8 macropore absorption Chromatography, 1 X 007 positive ion exchange chromatography,the result showed the purification product has prominent bioactivity inhibit Staphalococcus aureus.
