Prostaglandin E synthase (PGES) is the terminal rate-limiting enzyme for PGE2 synthesis,which catalyzes the conversion of PGH2 into PGE2. There are two isoforms of PGES,microsomal PGES(mPGES) and cytosolic PGES(cPGES). mPGES has been proved to be important for mouse implantation and bovine ovulatory process. The aim of this study was to examine the expression of mPGES and cPGES in mouse ovary during the sexual maturation by immunohistochemistry and in situ hybridization.mPGES mRNA was expressed in granulosa cells in mouse ovary during the sexual maturation,and the expression became stronger as the follicles developed. mPGES immunostaining was strongly detected on day 5 after birth. There was detectable mPGES immunostaining in the granulosa cells of the different follicles. The strongest mPGES immunostaining was seen in the primordial follicles,while a strong level of the immunostaining was also observed in the primary follicles,the secondary follicles,and mature follicles. The expression of cPGES mRNA in granulosa cells was stronger on day 1 after birth,and the lowest staining was detected in oocyte cytoplasm on day 5 after birth. The highest level of cPGES mRNA expression was seen in the granulosa cells and oocyte cytoplasm on day 10 after birth. But on day 15. the signal in the granulosa cells and oocyte cytoplasm became weak. Thereafter,the expression in the granulosa cells became stronger,while the expression in oocyte cytoplasm was low. There was no detectable cPGES immunostaining in the granulosa cells during the sexual maturation. A lowlevel of cPGES immunostaining was detected in oocyte cytoplasm on day 5 after birth. On day 10 after birth,the staining in oocyte cytoplasm was stronger,while the staining became weaker again on day 15. The strongest cPGES immunostaining was seen on day 20. Then cPGES immunostaining gradually decreased and reached to a basal level in adult ovary.
