The purpose of this paper is to identify sex of seedling Pistacia Vera. This study is helpful to Pistacia Vera tissue culture, rational field planting ratio of male to female Pistacia Vera in orchard for commercial production and lays a foundation for breeding of Pistacia Vera. The difference of male and female Pistacia Vera were compared and analyzed through treating with chemical reagent, surveying the morphology, RAPD techniques, analyzing the chromosome and comparing the band pattern of isoenzyme. The following results and conclusions were drawn: (l)The leaves of male and female Pistacia Vera were treated respectively with chemical reagent. The sexes were identified quickly and efficiently through different color and precipitate reaction among them. The difference is significant.(2)Some specific nucleoside acid bands were amplified from genomic DNA of male and female plants with RAPD technique, indicating that some gene marks related to sexes of Pistacia Vera were found. RAPD signal is an reliable, accurate identifying technique on sexidentification of Pistacia Vera. All these laid a foundation for sapling sex identification of this species by RAPD techniques. (3)The statistical analysis demonstrated that the foliar form exponent between male and female Pistacia Vera had marked difference. The analysis of the foliar form exponent can be used to identify sex of Pistacia Vera as a assistant means.Moreover, the sex difference of male and female Pistacia Vera can be clearly identified through observing chromosome and analyzing karyotype, but the operation of this technique is difficult. The results of this experiment are not perfect for various limiting factors. This study need to ulteriorly explore.
