High frequency regeneration of Gucumismelo L. cv Hetao occurred through a directly organogenetic developmental pathway in my experiment. The synthesized plant defensins gene RsFP2(R39) was cloned into plasmid vector pLJC18, then this fragment was inserted downstream of CaMV 35S promoter and enhancer ?Q ?of TMV in the binary vector pBJN438, named pBINYX. The cotyledons of Gucumis ,ne]o L. cv Hetao were transfered with A. tumnefaciens LBA4404 labering the expression vector pBTNYX, then induced resistant buds and get regeneration plants. PCR analysis showed that Rs- FP2(R39) gene has inserted into genomes of Cucwnis ,ne]o L.cv Hetao.
