The ITS-RFLP and electrophoretic karyotypes of Fusarium section Liseola were studied in the thesis. Five parts were studied: (1) ITS-RFLP of the standard isolates of six mating population in Fusarium section Liseola; (2) ITS-RFLP of the isolates of different mating type of mating population A in Fusarium section Liseola; (3) ITS-RFLP of the progeny of two isolates of mating populations A in Fusarium section Liseola; (4) the electrophoretic karyotype of the isolates of mating population A in Fusarium section Liseola; (5) The detectiOn of the colour and growth rate of the progeny of isolates of mating population A. On ITS-RFLP, a pair of oligonucleotide primers ITSI and ITS4 were used to amplify internal transcribed spacers (ITS) region of ribosomal DNA (rDNA) of Fusarium section Liseola.The amplified ITS fragments were about 620bp. The amplified ITS region was digested with restriction endonucleases Alu I ,Msp I and Taq I , the reaction tempreture were seperated by electrophoresis on 3% agrose gel. Popgene version 1 .31 was used to analyzed the bands of the pattern. The results of the study were as follows: RFLP were existed in the 12 standard isolates of 6 mating population in Fusarium Section Liseola. The isolates tested were clustered into two groups. all the standard isolates, except the mating population F, of the same mating population were divided into the same group, and the similarity level is above 78.3%. The different mating type of mating population were divided into two groups, and the similarity of only 45.5%. RFLP were also existed in the isolates of different mating type, the isolates tested were clustered into two groups. The similarity among the positive (+) mating type were very high, it 100%; the similarity among negative (-) mating type were 34.8-100%. On ITS-RFLP of the progeny of mating population A in Fusarium Section Liesola, the asexual progeny had the same restriction patterns with their own parent, but the sexual progeny had the same pattern with the maternity. On the electrophoretic karyotype (EK), a pair of isolates which belonged to mating population A of Fusarium Section Liseola, were chosen because of its good electrophoretic karyotype. Both strains were with 6 chromosomal DNAs, the estimated chromosomal DNA size of strain were 0.5Mb, 2.53Mb, 3.25Mb, 3.43Mb, 3.80Mb, 4.60Mb, and the asexual and sexual progeny isolates of Fusarium moniliforme were obtained. Both of the parental isolates were with 6 chromosomal DNAs. The estimated chromosomal DNA sizes of strain D47 were 0.5Mb 2.53Mb, 3.25Mb, 3.43Mb, 3.80Mb, 4.60Mb and strain D52 were 1.23Mb, 3.15Mb, 3.37Mb, 3.65Mb, 4.15Mb, 4.60Mb. The genome sizes of strain D47 and strain D52 were estimated to be 18.16Mb and 20.15Mb respectively. The progeny of D47 and D52 were also used to study the inheretance of Fusarium monil forme. Thc EK of the asexual progeny were the same with their parent, and the sexual progeny, just as the results of ITS-RFLP, were the same with their maternity.
