The research of distribution functions on quarks and gluons in nuclear is the most active frontier in nuclear and particle physics. EMC reflects that in heavy nuclei the reduce of high momentum (0.31 are necessary to illuminate the presence of short-range structure in nuclei.In this paper we used the convolution formula and considered the short-range corrections in nuclei and calculated the nuclear structure function on DIS of Lepton and helium in the large x region. From these results of calculation we can find that the distribution of nucleons are bell shaped with z, in the region of z>1, which the contribution of high momentum compositions in the nuclear spectral function can play a main role and the distribution of nucleons will be decreased with z. The nuclear structure function of helium will be decreased with the Bjorken scaling variable x. The calculation results showed that there is a consistency between the measurements of experimental data and the nuclear structure function. But overall, the nuclear structure function of helium will be larger than the carbon's, since in the large x region, especially in the region of x>1 for there will be a short range correlations in nuclei.
