Synaptotagmin genes which exsit as a large family are one kind of Ca2+.sensor that regulate the menbrane fusion in synaptic vesicle exo/endocytosis or neurotransmitter release process. All syts share a common structure namely an N-terminal transmenbrane sequence jointed to a linker of viarable length, followed by two tandemly arranged, distinct C2 domains which louse around in cytoplasm. Genes with the similar sequences as well as domain architecture to C2 domains were also found in plant genomes. There is a collection of six synaptotagmin-like genes in Arabidopsis. SytA has been reported to function in maintaining menbrane integrity or menbrane resealing from injury and regulate the endocytosis happened in virus movement proteins cell-cell transport. We study the localization of sytB on tissue level and its functions in vivo in this research.Here we generated the sytBpromoter:GUS transgenic plants, and then we performed GUS staining on different tissues. The GUS expression parttern is analyzed. To investgate into the functions of sytB gene in vivo, we bought three different T-DNA insertion lines as well as generated the overexpression lines and the homozygotes of them were used to study the phenotypes. Dataes indicated sytB has a strong expression in vascular system and mature pollens. The pollens of T-DNA lines germinated shorter pollen tubes than pollens of wild type did,however, the pollen germination rate and the growth of siliques didn't affected by the missing of sytB. Therefore, we suppose sytB might participate in regulating menbrane fusion in the tip growth of pollen tube, and also plays a role during the transportation of water, inorganic salt, nutrients or some other molecules through the vascular system.
