Low temperature stress is an important factor for abiotic stress and have a major impact on plant growth and development, geographical distribution and quality of production, etc. Low temperature also affect the activities of Nostoc flagelliforme in the process of life is an important limiting factor. As a low specificity Terrestrial cyanobacteria, the molecular mechanism of the individual growth of Nostoc flagelliforme has not yet proven, so far Nostoc or Nostoc cells in large-scale cultivation did not have a breakthrough. The growth of low-temperature conditions on Nostoc flagelliforme is appropriate.And there is a certain life activities stresses role, which inhibit the physiological activity of Nostoc cells, thus affecting the extracellular polysaccharide and protein yield. In order to study the molecular mechanisms of growth and development of Nostoc flagelliforme and its stress response mechanisms, particularly the mechanism of resistance to low temperature and freezing, from the level of proteins to explore their differences in protein and gene expression is particularly necessary.1. In this study, the modified TCA method is to optimize the proteome two-dimensional electrophoresis technology. Nostoc flagelliforme by extraction of total protein, total protein solubility, tape and other aspects of the selection and optimization of electrophoresis, have to get highly repetitive, good resolution of protein patterns.2. Further treatment at 4℃for 72h, the extracted proteins of Nostoc flagelliforme were separated by two-dimensional electrophoresis of total protein.3. Nostoc flagelliforme in protein expression under low temperature spectrum compared by PDQuest software analysis found that there have been 104 different protein spots. Among them,42 protein spots are up-regulated in the low temperature treatment and 62 protein spots are down-regulated in the low-temperature treatment. 4. Initially screened significantly different part of the protein spots by mass spectrometry analysis of these proteins, including phytochrome 3, putative chloroplast phosphoglycerate kinase, H+- or Na+- translocating f-type,v-type and A-type ATPase superfamily,Plasma membrane associated proteins and other unknown proteins, etc., initially speculated that these differences may be related to low temperature stress proteins related.Conclusion:Application of 2-DE and MALDI-TOF-MS method, We have established a low-temperature stress Nostoc sp total protein two-dimensional gel electrophoresis, isolated and identified 23 species and differential expression of low temperature stress-related proteins. Associated with stress resistance by looking for these proteins (or genes), understanding the mechanisms of Nostoc algae and to enhance their resistance resilience of great significance, which also issued a state of low temperature stress the physiological mechanism of Nostoc A new clue.
