Antimicrobial peptides also known as antimicrobial peptides,peptide antibiotics, whitch first discovered by Swedish scientists Boman in 1980, and the cecropins purified by Hultlmark, in the following year its amino acid sequence published on"Nature".Antimicrobial peptides are found widely distributed in bacteria, plants, insects, fish, amphibians, mammals (including humans) . With broad-spectrum bactericidal, inhibiting the virus, such as inhibiting tumor cell killing activity, but also accelerate the immune and wound healing process.In this study, ethyl acetate extraction of the Yarkand from the injured skin Triplophysa extracted the protein (peptide), the extracted crude extracts after SephadexG-50 and 2 column and anion exchange adsorption column Purified twice.The purified peptide use Ninhydrin color reaction, and then detection by the Tricune-SDS-PAGE protein gel electrophoresis the molecular weight of peptide size 9700 Dalton, determined by Bradford peptide concentration: 0.38 mg/mL. Followed by antimicrobial peptides against Pathogenic E. coli, Staphylococcus aureus, fungus, Verticillium dahliae cotton antibacterial test,at the same time use of double dilution method to detect Triplophysa Yarkant antimicrobial peptides minimum inhibitory concentration 4μg/mL.Detection the antibacterial activity of antimicrobial peptides, and research the impact temperature, pH, light on activity of antimicrobial peptides , the results indicating that range of Yarkand Triplophysa antimicrobial peptides in 25-100℃, antioxidant activity did not change significant; Yarkand Triplophysa of antimicrobial peptides in pH = 7.0 and the strongest antioxidant activity. When the pH <7, with the pH ecrease, DPPH and ABTS+scavenging activity of free radicals decreased, but no significant change; when the pH> 7, with the pH increased, the removal of two radicals capacity has decreased significantly. Especially when the pH value increased to 10.0, the peptide DPPH and ABTS+free radical removal ability, lost more than 90% and 20%. Sunlight on the Yarkand Triplophysa antioxidant activity of antimicrobial peptides have some damaging effects, and indoor natural light and placed in the dark on its antioxidant activity was less affected.
