| Objective Irritable Bowel Syndrome(IBS)is a common functional gastrointestinal disease,which is characterized by abdominal discomfort and abdominal pain.According to the current reports,the incidence of IBS in China is about 6%,which ranges from 5% to 45% in the world.So far,the pathogenesis and mechanism of IBS have not been completely clear,and the clinical treatment is still symptomatic treatment.Although most patients do not develop life-threatening symptoms,chronic abdominal discomfort can have a significant impact on the quality of life,at the same time,the complexity of its pathogenesis leads to complicate and changeable clinical symptoms or recurrent attacks,which also causes huge life discomfort and economic burden to the patients.Periplaneta Americana is a genus of Periplaneta(INSECTA),which belongs to the family Blatttodea(Blatttidae).As a traditional Chinese medicine,Periplaneta Americana has the functions of dispersing silt,eliminating accumulation,detoxifying,promoting water retention and detumescence.It can be used to treat IBS in clinic with remarkable curative effect.In this study,network pharmacology was used to predict the pharmacological basis of Periplaneta Americana and reveal its target and signal pathway,and to establish mouse PI-IBS model,to clarify the mechanism of Periplaneta Americana in the treatment of IBS,and to verify the main pharmacodynamic material basis of Periplaneta Americana in the network pharmacology in the treatment of IBS,based on the above research results,the characteristic chromatogram and quality control methods were established to control the content of main active ingredients in order to ensure the curative effect.Therefore,the innovative establishment of Network Pharmacology and Intestinal Flora metabolism of the drug research model is expected to find drug action targets and pathways for researchers in the future,to study the mechanism of drug action and provide new research ideas and methods.Using HPLC technology to identify the species of Periplaneta americana with high specificity can also more effectively,quickly and accurately complete the accurate identification of traditional Chinese medicine,which provides a more powerful means for the quality control of traditional Chinese medicine,and provides innovative power and scientific basis for the development of traditional Chinese medicine industry and large variety cultivation.Method Firstly,based on literature review and previous studies,Amino acids and nucleosides,which are the main medicinal components of Periplaneta Americana,were selected,and the potential targets of Periplaneta Americana were predicted using Pharm Mapper database,the protein-protein interaction network was established with CYTOSCAPE3.7.1 software and enriched with David database and Kegg pathway.The network of Chemical constituents-target-pathway of Periplaneta Americana was established with CYTOSCAPE3.6.1 software,the target and pathway of IBS were analyzed,and the mechanism of IBS related target and pathway was elucidated from the perspective of network pharmacology.Secondly,the PI-IBS model of mice was established by gavage of Campylobacter Jejuni.After gavage of different dosages of Periplaneta Americana,the microbial diversity of prokaryotes in different groups was detected by 16 s r RNA amplification high-throughput sequencing technique The diversity of the samples was calculated by Chao1,Simpson,Shannon and Faith’s PD index to indicate the species richness of the intestinal microflora in different groups;the diversity of the samples was calculated by the nuweighted Unifrac distance matrix principal coordinate analysis to explain the variation of the samples in different groups;and the species information and abundance of the marker groups in different groups were analyzed by Random Forest Gini Analysis and index representation to evaluate their importance in intergroup classification.In order to elucidate the effect of Periplaneta Americana on the intestinal microecology of mice,to reveal the mechanism of its action in the treatment of IBS,to verify the results of network pharmacology,and to form the mechanism of the treatment of IBS of Periplaneta Americana based on the network pharmacology and the metabolism of intestinal flora.Finally,In order to control the quality of Chinese herbal medicine,We used HPLC fingerprint technology and content determination methods to control the quality of the pharmacodynamic substances,which was determined by the above studies.Results In this paper,we established an innovative method of Network Pharmacology and Intestinal Flora metabolism to elucidated the target and pathway of IBS treatment of Periplaneta Americana.Studies shows that Periplaneta Americana could regulate and rebuild intestinal flora to treat IBS,and through the above research,we has elucidated the pharmacodynamic material basis of Periplaneta Americana,and has established the quality control method,including HPLC technology and characteristic chromatogram of Periplaneta Americana.Conclusion To sum up,this paper has achieved good scientific research results,through the innovative establishment of Network Pharmacology and intestinal flora metabolism as a model,it provides a new idea and method for finding the target and pathway of drug action and studying the mechanism of drug action.Through the use of the model to determine the material basis of efficacy,and then to carry out quality control,it also achieves the goal of effectively ensuring the efficacy and quality stability of clinical medication,therefore,the application of this research idea and method in drug quality control is highly innovative and scientific,which leads to the sale and use of drugs and achieves high social and economic benefits,at the same time,it also plays a good demonstration role in guiding the cultivation of large varieties of Chinese medicinal materials.Some of the research results of this project have been published 2 SCI articles and 5 articles in Chinese core journals,which have won the first prize of Sichuan Science and technology progress. |
