| In this study,a detection method and an abuse purchasing method of 10 common illicit drugs in China were established.According to related information of drug abusers in Shenzhen,Guangdong,Heroin,Ice,Ketamine and Mehtadone were found to be the 4 kinds of most populated drugs in Shenzhen.A comparasion and relationship of these drugs’concentration in the 5 different areas of head vertex were performed,and metabolomics studies were performed to evaluate the metabolites in hair of heroin and ice abusers.1.The method for the determination of 10 kind drugs in vivo was established.These 10 drugs include heroin,morphine,codeine,ice,amphetamine,ecstasy,cocaine,phencyclidine,ketamine and methadone.The analyses were performed on UPLC-MS/MS.Liquid chromatographic separation was achieved on a Agilent Zorbax ECLIPSE Plus C18 chromatography column,using 0.1%FA-ACN and 0.1%FA-H2O as mobile phase with gradient elution.Mass spectrometry was equipped with ESI+ ion source in multiple reaction monitoring and electro spray voltage was set at 3.0 kV.Whole analysis could be finished in 6 min.The method meets the requirement for method validation,and could accurately detect the concentration during the range of 2~1000 ng·mL-1.500 hair samples collected from vertex posterior were analysised,and the method was evaluated by detection of GFTCH profiling samples and comparing results of urine tests.The results showed that this method could accurately detect concentration of 10 drugs.In addition,4 kinds of the most abused drugs were found as heroin,ice,ketamine and methadone.2.A method was devloped for purchasing abuse history.16 hair samples were collected from abusers,who have long abuse history and hair length longer than 12 cm.Hair samples were cut 12 cm from scalp,and cut into 12 aliquots.Method of purchasing the history was established and meets the requirement,and applied to the determination of all the samples.Results showed that this method could precisely purchase the abuse history in the time accuracy of month.The concentration and relationship of the 4 kinds of most abused drugs among 5 areas in scalp have been studied.Results showed that concentration in these 5 areas have significant correlation,though different drugs have different concentrations.Heroin has a biggest concentration in vertex anterior,ketamine,ice and methadone all have the biggest concentration in lateral right.3.Heroin abusers’ hair specimens were used for the study of the influence of heroin to in vivo metabolites by metabolomics technology.58 and 72 hair samples of heroin users and non-heroin users have been collected.The analyses were performed on UFLC-IT-TOF.Liquid chromatographic separation was achieved on a Aglient Zorbax ECLIPSE Plus C18 UPLC coloum with ACN-0.1%FA-H2O as mobile phase.Ion source of mass spectrometry was ESI+/-.The mass parameters were set as follwing:interface voltage 4.5 kV,heatblock 200℃,CDL 200℃,N2 gas folwrate 1.5 L/min,CID energy 30%,TOF dector voltage 1.6 kV.Analysis could be finished in 20 min.Samples were classified by PLS-DA analysis,and 2 primary compontents explained 78%varieties.Then biomarkers were identified.Sorbitol,Myristic acid,Linoleic acid,Arachidonic acid,Glutathione and Cortisol were identified as biomarker,and compared with blank group,concentration of Sorbitol and Cortisol have increase trends,Myristic acid,Linoleic acid,Arachidonic acid and Glutathione have decrease trends.4.Ice abusers’ hair specimens were used for the study of the influence of heroin to in vivo metabolites by metabolomics technology.80 and 44 hair samples of heroin users and non-ice users have been collected,And were analysis by the established method for herioin abusers.Samples were classified by PLS-DA analysis,and 2 primary compontents explained 77%varieties.And then biomarkers were identified,and Glutamate,Homocysteic acid,Glutathione,Prostaglandin F1αAlcohol,Cortisol,6-keto-Prostaglandin F1α,Lysophosphatidylcholine(18:2),etc,those 8 compounds were identified as biomarker,and compared with blank group,concentration of Glutamate,Homocysteic acid and Cortisol have increase trends,Glutathione,Prostaglandin F1αAlcohol,6-keto-Prostaglandin Fla and Lysophosphatidylcholine(18:2)have decrease trends. |
