Study On The Inhibitory Effects Of Electrospinning Materials On Oral Microbial Biofilm Formation

Abstract/Summary:
Objectives: The microorganisms represented by Streptococcus mutans in the oral cavity are important pathogenic factors of oral infectious diseases.Most of these microorganisms need to adhere to the surface of the oral medium and form a biofilm to express pathogenicity.Electrospinning material is a new type of organic polymer material.Past studies have mostly focused on its functions such as tissue engineering and drug loading,but there are few studies on its antimicrobial adhesion.This research focused on the phenomenon of electrospinning materials in inhibiting the adhesion of common pathogenic microorganisms in the oral cavity and discovered the mechanism of inhibiting adhesion.At the same time,it explored its biological safety and preliminary exploration of its potential application mode.Methods:1.Prepared and improved the electrospinning material using polystyrene(PS)as raw material;observed that the electrospinning material inhibits common oral cavity by crystal violet staining,anthrone-sulfuric method,scanning electron microscope,laser confocal microscope,etc.Adhesion of pathogenic bacteria(Streptococcus mutans,Porphyromonas gingivalis,Staphylococcus aureus,Pseudomonas aeruginosa,etc.);the biofilm adhesion of the two species of Streptococcus mutans and Candida albicans was observed.2.The effect of electrospinning raw materials and spatial structure on the adhesion of Streptococcus mutans was explored;the surface charge properties of electrospinning materials were characterized by Zeta potential,and the surface charge properties of electrospinning materials were explored by scanning electron microscopy and CFU colony counting.For the influence of the adhesion ability of Streptococcus mutans;by simulating changes in the oral environment(calcium ion,iron ion concentration,p H,sucrose concentration,etc.),the ability of electrospinning materials to inhibit the adhesion of Streptococcus mutans under different environments is explored.3.Through the cytotoxicity experiment CCK8,we explored the cytotoxicity of electrospinning materials to HGE cells and HOK cells;explored the ability of electrospinning materials to carry calcium fluoride nanoparticles and release fluoride ions;explored the effects of electrospinning materials and The binding ability of orthodontic retainer materials;an improved electrospun material retainer was made and placed on an isolated tooth model to simulate wearing the retainer in vitro;bacteria on the surface of the retainer were collected and explored by scanning electron microscopy and CFU colony counting Anti-adhesion effect of Streptococcus mutans.Results:1.Successfully manufactured Polystyrene/Polyvinylpyrrolidone(PS/PVP)electrospinning materials;found that PS/PVP electrospinning materials have effectively inhibited the adhension of Streptococcus mutans,Porphyromonas gingivalis,Staphylococcus aureus and Pseudomonas aeruginosa.Besides,it reduced the adhesion of the biofilm formed by the double species of Streptococcus mutans and Candida albicans,and this reduction was more obvious for Streptococcus mutans.2.The raw material of PS/PVP itself has a certain degree of anti-streptococcus mutans adhesion effect;the PVP component does not affect the anti-streptococcus mutans adhesion ability of the electrospinning material;Zeta potential shows that PS and PS/PVP with negative surface potential have the ability to resist the adhesion of Streptococcus mutans,and this ability is related to the amount of surface charge;changing the concentration of calcium ions and iron ions,the p H of the medium,and the concentration of sucrose in the medium will not affect the PS/PVP electrospinning The ability of the material to resist the adhesion of Streptococcus mutans proves that this material has certain environmental resistance.3.Both HGE cells and HOK cells cultured on PS/PVP electrospinning materials grew well,which proved the biological safety of the materials;calcium fluoride nanoparticles immobilized on PS/PVP electrospinning materials were observed,The drug release experiment proved that it can slowly release fluoride ions;the holder plastic combined with the electrospinning material can be normally prepared into an orthodontic holder;the electrospinning material with fixed calcium fluoride particles improves the variation chain on the surface of the holder The amount of cocci has been significantly reduced.Conclusions: This study found that PS/PVP electrospun material inhibited the adhesion of oral pathogenic microorganisms and explored its mechanism of inhibiting the adhesion of Streptococcus mutans.The electrospinning material improved orthodontic retainer can slowly release fluoride ions,which provides a new idea for the prevention of orthodontic caries and has clinical significance.
