PurposeStomach cancer is a globally important disease,the fifth most common diagnosed malignant disease,and the third most common cause of cancer death in the world.It has a high incidence in high-risk Asian regions.Since it is often diagnosed in advanced stages,the death rate associated with gastric cancer is higher.Advanced gastric cancer is characterized by extensive morphological diversity.According to histological classification,gastric adenocarcinoma is the most common histopathological classification.In recent years,researchers have tried to link molecular characteristics with histological phenotypes and clinical characteristics.Some previous studies have shown that LncRNAs have a key regulatory role in the biology of gastric cancer,and the mechanism of their role in gastric cancer is still very important.To a large extent unknown.In this study,LncRNA BNC2-AS1,a newly identified non-coding RNA,was mainly discussed in terms of its expression in gastric adenocarcinoma tissues and gastric adenocarcinoma cells,from the molecular characteristics of gastric cancer and its histological phenotype.Further explore the mechanism of LncRNA BNC2-AS1 on the biological function of gastric cancer.Therefore,this study identified a new clinically useful monitoring marker and a potential new molecular therapeutic target that can guide clinical treatment in order to guide the future The development of treatment methods.MethodIn 106 pairs of gastric adenocarcinoma and its corresponding adjacent tissue specimens,real-time fluorescent quantitative PCR(RT-q PCR)was used to verify the expression level of lnc RNA BNC2-AS1.By comparing the expression and survival prediction of BNC2-AS1 in the Cancer Genome Atlas(TCGA)/genotype-tissue expression(GTEx)database,the comprehensive evaluation result is consistent with the expression profile.(2)In gastric adenocarcinoma cell lines,the expression level of lnc RNA BNC2-AS1 detected by q RT-PCR method.The effect of lnc RNA BNC2-AS1 on the biological behavior of gastric adenocarcinoma cell expansion,invasion and differentiation was evaluated by cell function assay.(3)Predict the downstream target genes of BNC2-AS1 through the prediction website Starbase.By inhibiting the expression of downstream target genes,explore the role of lnc RNA BNC2-AS1 and the corresponding target genes and the influence of the involved tumor network pathways on the molecular characteristics of gastric cancer cells.Result(1)Comparing gastric cancer tissue with its neighboring tissues,the expression level of LncRNA BNC2-AS1 showed high expression in gastric cancer specimens(P<0.05),with an up-regulation rate of 58.5%;in gastric cancer tissues,the expression of LncRNA BNC2-AS1 The level of the tumor is positively correlated with the size of the tumor,the degree of progression,the presence of distant metastases,blood vessels,nerves,and lymph nodes(P<0.05);the expression level of LncRNA BNC2-AS1 is inconsistent with the database results,which may be due to excessive sample size LncRNA BNC2-AS1 high expression prediction may affect the prognosis of patients with advanced gastric cancer;(2)In gastric cancer cell lines,LncRNA BNC2-AS1 showed an up-regulated expression level(P<0.05).By reducing the expression of LncRNA BNC2-AS1,it was found that the expansion,migration,and invasion ability of gastric cancer cells was inhibited(P<0.05);(3)In gastric cancer cell lines,LncRNA NCL showed high expression(P<0.05);Reducing the expression of LncRNA NCL can inhibit the expansion,apoptosis and migration of gastric adenocarcinoma cells(P<0.05).ConclusionThrough q RT-PCR,it was verified in 106 fresh gastric cancer tissues and adjacent normal tissues after surgery.In gastric cancer tissues,LncRNA BNC2-AS1 showed an up-regulated expression level,which was consistent with the expression profile results.In gastric cancer tissues and matched adjacent tissues,the differential expression of LncRNA BNC2-AS1 is closely related to the tumor size,distant organ metastasis,blood vessel,nerve,and lymph node invasion in patients with gastric adenocarcinoma.We have studied the function of gastric cancer cells and found that inhibiting the expression of LncRNA BNC2-AS1 can significantly affect the proliferation,migration and invasion of gastric cancer cells and other molecular biological behaviors.In summary,LncRNA BNC2-AS1 is expected to become a useful monitoring marker and a potential new molecular therapeutic target that can guide clinical work.
